Enantioselective hydrogenation of -aminomethylacrylates containing a free NOH group for the synthesis of -amino acid derivatives

چکیده

We describe highly enantioselective synthesis of -amino acid derivatives (1a-c) using asymmetric hydrogenation of -aminomethylacrylates (2a-c), which contain a free basic NOH group, as the key step. The -aminomethylacrylates (2a-c) were prepared using the Baylis– Hillman reaction of an appropriate aldehyde with methyl acrylate followed by acetylation of the resulting allylic alcohols (4a-b) and SN2 -type amination of the allylic acetates (3a-b).

Hydrogenation of a series of cyclic beta-(acylamino)acrylates with tetrasubstituted olefins has been accomplished successfully with the use of Ru catalysts with chiral biaryl ligands such as C3-TunaPhos, and up to over 99% ee's have been achieved.
